I came across a PCR kit ("Invitrogen™ Platinum™ SuperFi II DNA Polymerase") that claims to enable universal primer annealing at 60°C. I'm curious about how this is achieved.
What mechanisms or additives allow them to stabilize primer annealing at this temperature regardless of whether my primer normally anneals at a lower (e.g., 50°C) or higher (e.g., 72°C) temperature?How do they balance the melting temperature differences?
